DNA Walker_lineW（for detection of N gene of SARS-CoV-2)
BBa_K3480002, what we called DW_lineW, is an oligonucleotide which used to build a nano machine called DNA Walker. DW_lineW is used to identify and bind potential viral genomes in the environment, thus promote the working of DNA Walker. Unlike DW_lineL, lineW is designed hang out of the DNA Walker machine，and becomes part of the machine by pairing with the 5' half of target sequence, which pairing with lineL at the other half sequence. DW_lineW has 15 bases at the 5' end which are paired with a conserved site in the N gene of SARS-CoV-2. Then there is a reigon of oligoT(30nt) which can promote the flexibility of DW_lineW and reduce steric hindrance. DW_lineW has 19 bases at the 3' end whose sequence is complementary to 3' end of DW_lineC. DW_lineW can only work normally in DNA Walker. DW_lineW and DW_lineL will identify the 3' half and 5' half of the target sequence respectively.
